Research IDs potential treatment for deadly, HIV-related blood cancer

June 25, 2013 ? Researchers at the USC Norris Comprehensive Cancer Center have discovered a promising new way to treat a rare and aggressive blood cancer most commonly found in people infected with HIV.

The USC team shows that a class of drugs called BET bromodomain inhibitors effectively targets primary effusion lymphoma (PEL), a type of cancer for which those drugs were not expected to be effective.

"It's a reversal of the paradigm," said Preet Chaudhary, chief of the Nohl Division of Hematology and Blood Diseases at the Keck School of Medicine of USC and principal investigator of the study. "Our results suggest that this new class of drug may be an effective treatment for a wider range of cancers than previously thought."

PEL is caused by infection with Kaposi's sarcoma-associated herpes virus, the most common cause of cancer among patients with AIDS. The prognosis for PEL is poor, with a median survival of three to six months. Thus, there is a critical need for new therapies for the disease.

Chaudhary and his colleagues show that inhibitors targeting the BRD4 protein blocked growth of PEL cells in a test tube and in a mouse model. The results were surprising because BET inhibitors were thought to be only effective against cancers linked to an overexpression of the Myc gene.

"We actually found that cancers that overexpress Myc are not as responsive to BRD4 inhibitors. PEL is more responsive," Chaudhary said.

Cancers like multiple myeloma and Burkitt's lymphoma overexpress the Myc gene and have been shown to respond to BRD4 inhibitors. In PEL, the Myc gene is moderately expressed and there is no chromosomal translocation as is seen in multiple myeloma or Burkitt's.

More research is needed to create compounds ready for testing in people. Once those drugs are ready for clinical trial, data from this study suggest that they may treat a wide range of cancers. Chaudhary anticipates testing them alone and in combination with other drugs.

VTT Technical Research Centre of Finland has developed a fast and affordable testing device for detecting the presence of toxic blue-green algae in water. There is currently no fast, affordable and user-friendly way for consumers to check water quality themselves.

The blue-green algae testing kit developed by VTT and the University of Helsinki is like a miniature laboratory. The device is the size of a thermometer, and it contains antibodies that react to any toxic bacteria found in a water sample. The test reveals in minutes whether the water sample contains toxic blue-green algae.

Thanks to the new testing device, consumers will soon be able to check themselves whether water is safe for swimming. At the moment, information on blue-green algal blooms in water is mostly based on visual inspections. However, visual inspections alone are not capable of determining whether an algal bloom is toxic. Until now, the toxicity of algae has generally had to be tested in a laboratory. For example, only approximately half of blue-green algal blooms in lakes are toxic and harmful to humans and animals. The new testing kit provides a fast and reliable means of determining whether a blue-green algal bloom is toxic.

Blue-green algae, also known as cyanobacteria, favour eutrophic and warm water. Cyanobacteria can be found in almost every terrestrial and aquatic habitat - oceans, fresh water, damp soil, temporarily moistened rocks in deserts, and even Antarctic rocks. Every year, they form extensive blooms e.g. in the Baltic Sea and other waters. The prevalence of algae each summer depends on factors such as weather and water nutrient levels. The first blue-green algal blooms begin to form when the surface of sea water reaches 15 degrees.

The testing kit for detecting toxic blue-green algae is in the process of being commercialised. The kits could be on sale within 23 years.

It's an odd choice that's unlikely to much affect Kanye West's and Kardashian's little girl ? but, for a child born to non-famous parents, is a name that might critically shape who she grows up to be. Without the gilded Kardashian name to guarantee her success, that non-celebrity girl might struggle to fend off bullies, get hired, and overall surmount other people?s ? and eventually her own ? low expectations for her future.

Studies have increasingly shown that names are a highly relevant factor is how others perceive us and we perceive ourselves. In 2010, David Figlio of Northwestern University in Illinois analyzed names from millions of birth certificates for the probability that the name belonged to someone of low socioeconomic status ? children whose names met those criteria would go to be discriminated against throughout life, he found. Similarly, a 2003 study from The National Bureau of Economic Research found that resumes with White-sounding names receive 50 percent more callbacks for interviews than resumes with African-American-sounding names.

The significance of that research has grown in recent years, as baby names have become increasingly more unusual. In 2010, a British study of some 3,000 parents found that one-in-five of them regretted the name they had selected for their children, in that case often an unusual name or one with a strange spelling. That finding wasn?t surprising to scientists, since a growing crop of studies have linked unusual names to numerous disadvantages in life.

Much of how we perceive the world is unconscious, and our latent biases against particular names are often influential in how we treat people. A 2011 informal survey that combed baby name conversations on online message boards found that the names perceived to be highly trendy are the biggest culprits in jolting those biases and that those names often end up capping our lists of the most hated names.
